I was going to list my favorites from Buffy, and then I realized that I pretty much listed everyone but Tara, Dawn and Riley.

Also need to mention Lorelai Gilmore. If the world was full of Gilmores, it would really be a better place.

Scooby Doo, Shaggy, and Fiona from So Weird get honorable mentions.

Since I'm guessing nobody on this board watches this show, I'll say Omar from "The Wire." Feared, highly intelligent and somewhat crazy gay (lol!) leader of a gang of drug dealer robbers. Kinda a sub character on the show, but definitely one that shines. Probably one of the more unique characters in recent TV history. I'd love to see the actor that plays him do more stuff.
